What is the role of eIF2B in the Integrated Stress Response (ISR)?
eIF2B is a protein complex found in eukaryotes. It is the guanine nucleotide exchange factor for the eukaryotic initiation factor 2 and therefore regulates the initiation of protein translation.
How does a persistent ISR lead to cellular dysfunction?
The ISR is a fundamental
process that is transiently induced to help cells adapt to various types of
stress . When
persistently active, the ISR may be harmful to cells.
Targeting the ISR in ALS
The ISR is activated in people with ALS causing a reduction in normal protein synthesis, increased stress protein production, and formation of stress granules containing TDP-43. Treatments are available that can dissolve these stress granules in motor neurons.
